01Research context
CJC-1295 (No DAC) and Ipamorelin act on two different receptors of the same endocrine axis and are commonly combined in preclinical research because their effects on pituitary somatotrophs are complementary. CJC-1295 (No DAC) binds the GHRH receptor and raises the amplitude of growth-hormone pulses; lacking the Drug Affinity Complex, it does not bind serum albumin, so it has a short half-life and preserves the natural pulsatile release pattern rather than producing sustained elevation. Ipamorelin initiates a growth-hormone pulse through the ghrelin receptor and, relative to earlier secretagogues such as GHRP-2 and GHRP-6, shows minimal stimulation of cortisol, prolactin, or ACTH in research models. Because the two pathways converge on somatotroph cAMP and calcium signalling, co-administration in animal models produces a greater GH response than either peptide alone, making the pair a standard tool for pulse-amplitude, pulse-frequency, IGF-1 axis, and comparative secretagogue-selectivity studies.
